Description
Indications
Meclizine Hydrochloride and Pyridoxine Hydrochloride are used for the prevention and treatment of nausea, vomiting, dizziness, and motion sickness. They are also effective in managing radiation sickness and vertigo associated with vestibular system disorders, such as Meniere’s syndrome, labyrinthitis, and other balance-related disturbances.

Pharmacology
Meclizine is a piperazine-derived antihistamine with antiemetic, anticholinergic, and antihistaminic properties. It works by reducing the sensitivity of the inner ear (labyrinthine apparatus) and may act through nerve pathways that connect to the vomiting center (VC) from the chemoreceptor trigger zone (CTZ) or other central nervous system (CNS) pathways.
Pyridoxine (Vitamin B6) is included in the formulation to enhance the antiemetic effect and also serves as a dietary supplement.
Dosage & Administration
Adults and children aged 12 years and older:
- Nausea and vomiting: 25-50 mg once daily or as prescribed by a physician.
- Motion sickness: Take 25-50 mg at least one hour before travel. The dose may be repeated every 24 hours as needed during the journey.
- Radiation sickness: 50 mg should be taken 2 to 12 hours before radiation therapy.
- Vertigo: 25-100 mg per day in divided doses, as advised by a physician.
- Prevention of nausea and vomiting associated with emergency contraceptive pills (ECP): 25-50 mg should be taken one hour before the first ECP dose. If necessary, another dose can be taken 24 hours later.
The safety and efficacy of this medication in children under 12 years of age have not been established.

Drug Interactions
- The sedative effects of Meclizine may be enhanced when taken with alcohol or other central nervous system (CNS) depressants, including benzodiazepines, barbiturates, tricyclic antidepressants, opioids, skeletal muscle relaxants, and other antihistamines.
- Co-administration with other anticholinergic drugs may intensify anticholinergic effects.
- Meclizine can slow gastrointestinal motility, leading to increased absorption of digoxin.
Contraindications
This medication is contraindicated in individuals with known hypersensitivity to Meclizine Hydrochloride or Pyridoxine Hydrochloride.

Common side effects may include:
- Drowsiness
- Dry mouth
- Occasionally, blurred vision
Pregnancy & Lactation
- Pregnancy Category B: Large-scale studies have not shown harmful effects on the fetus. Available data suggest that Meclizine poses minimal risk of birth defects and is considered a first-choice treatment for nausea and vomiting during pregnancy.
- The safety of Meclizine in breastfeeding women has not been established. Consult a healthcare professional before use.
Precautions & Warnings
- Meclizine Hydrochloride may impair mental alertness and physical coordination, potentially affecting activities such as driving or operating machinery.
- Alcohol consumption should be avoided while taking this medication.
- Due to its anticholinergic properties, use with caution in patients with asthma, glaucoma, or an enlarged prostate gland.
Overdose Effects
- Symptoms: Severe excitability, seizures, drowsiness, and hallucinations.
- Treatment: Supportive and symptomatic care should be provided. Dialysis may be considered if necessary.
