Description
Afun is a broad-spectrum antifungal medication used to treat various skin infections caused by pathogenic dermatophytes, yeasts, and molds. Additionally, it has activity against certain gram-positive bacteria, including Staphylococci and Trichomonas.
Similar to other imidazole antifungals, Afun works by disrupting the permeability of fungal cell membranes. At lower concentrations, it inhibits ergosterol synthesis, an essential component of fungal cell walls. At higher concentrations, it causes direct membrane damage, leading to fungal cell death.
Pharmacology
Clotrimazole, the active ingredient in Afun, functions by damaging the fungal cell membrane’s permeability barrier. It inhibits ergosterol biosynthesis, which is crucial for maintaining the integrity and function of fungal cell walls. Without sufficient ergosterol, fungal cells cannot sustain their structure, leading to inhibited growth.
Beyond its antifungal effects, Clotrimazole also impacts other biological mechanisms, such as:
- Inhibiting sarcoplasmic reticulum Ca²⁺ ATPase
- Depleting intracellular calcium levels
- Blocking calcium-dependent potassium and voltage-gated calcium channels
These additional effects contribute to Clotrimazole’s pharmacological profile beyond its primary antifungal properties.
Dosage & Administration
Clotrimazole Cream
- Apply a thin layer to the affected area 2-3 times daily, rubbing it in gently.
- Due to its high efficacy, a small amount is sufficient for an area about the size of the palm.
- Consistent and prolonged application is crucial for successful treatment.
Recommended Duration of Treatment:
- Dermatomycoses: 3-4 weeks
- Candida vulvitis and Candida balanitis: 1-2 weeks
- Erythrasma and Pityriasis Versicolor: Approximately 3 weeks
- For fungal foot infections: Treatment should continue for 2 weeks beyond the disappearance of symptoms to prevent recurrence.
After washing, the feet should be thoroughly dried, particularly between the toes. Clotrimazole Cream is odorless, non-staining, and can be washed off easily.
Clotrimazole Topical Solution
- Apply sparingly to the affected area 2-3 times daily and rub in gently.
