Anadol SR 100 mg
Description
Indications
Anadol SR is prescribed for the management of moderate to severe pain. It is commonly used for conditions such as:
- Postoperative pain
- Colic and spastic pain
- Cancer-related pain
- Joint pain
- Neck and back pain
- Pain associated with osteoporosis
Use only as directed by a registered physician.
Pharmacology
Tramadol, the active ingredient in Anadol SR, is a centrally acting synthetic analgesic. It works by inhibiting the reuptake of serotonin and noradrenaline, thereby modifying pain signal transmission. The analgesic effect of Tramadol is mediated through stimulation of mu-opioid receptors and indirect modulation of central monoaminergic inhibitory pathways.
Dosage & Administration
Capsule or Tablet:
- The usual dose is 50 to 100 mg every 4 to 6 hours.
- For acute pain, an initial dose of 100 mg is recommended.
- For chronic pain, an initial dose of 50 mg is advised, followed by 50 to 100 mg every 4 to 6 hours as needed.
- The maximum daily oral dose should not exceed 400 mg.
Sustained Release (SR) Capsule or Tablet:
- Take one SR capsule or tablet every 12 hours (e.g., morning and evening).
- The number of capsules per dose depends on pain severity but should not be taken more frequently than every 12 hours.
- The total daily dose should not exceed 400 mg.
Injection:
- A dose of 50-100 mg may be administered every 4 to 6 hours via intramuscular injection or intravenous infusion.
- For postoperative pain, an initial dose of 100 mg is followed by 50 mg every 10 to 20 minutes if needed, with a maximum of 250 mg in the first hour.
- Thereafter, doses of 50 to 100 mg can be given every 4 to 6 hours, with a total daily dose not exceeding 600 mg.
Suppository:
- Tramadol suppositories should be administered rectally.
- The usual adult dose is 100 mg every 6 hours.
- A total daily dose of 400 mg (four 100 mg suppositories) is typically sufficient.
- In cases of cancer pain or severe postoperative pain, higher daily doses may be required.

Drug Interactions
- Anadol SR generally has minimal drug interactions.
- However, it should not be combined with monoamine oxidase (MAO) inhibitors.
- Concurrent use of carbamazepine may increase the metabolism of Anadol SR, requiring a higher dose for effectiveness.
Contraindications
Anadol SR is contraindicated in individuals with:
- Known hypersensitivity to Tramadol
- Acute intoxication due to alcohol, hypnotics, opioids, centrally acting analgesics, or psychotropic drugs
Side Effects
Common side effects include:
- Dizziness, nausea, vomiting
- Constipation, headache, drowsiness
- Pruritus, sweating, dry mouth, diarrhea
Less common side effects include:
- Allergic reactions, weight loss, abdominal pain
- Palpitations, vasodilation, gastrointestinal bleeding
- Stomatitis, hepatitis
Pregnancy & Lactation
- The safety of Tramadol use during pregnancy has not been established. It crosses the placenta and should only be used if the potential benefit outweighs the risk to the fetus.
- Tramadol and its metabolites are excreted in breast milk; therefore, it is not recommended for breastfeeding mothers.
Precautions & Warnings
- Respiratory depression: High doses or concurrent use with anesthetics or alcohol can cause respiratory depression.
- Opioid dependence: Anadol SR is not suitable for opioid-dependent patients.
- CNS depressants: Use with alcohol, opioids, anesthetics, tranquilizers, or sedatives requires caution and dose adjustments.
- MAO inhibitors: Anadol SR should be used with extreme caution in patients taking MAO inhibitors, as Tramadol affects norepinephrine and serotonin levels.
- Head injuries and intracranial pressure: Caution is advised in patients with head injuries or increased intracranial pressure.
- Acute abdominal conditions: Use with caution in patients with acute abdominal pain.
Use in Special Populations
- Children aged 1 year and above: Anadol SR may be given at a dose of 1-2 mg/kg body weight.
- Children under 14 years: The suppository form (100 mg) should not be used.
- Anadol SR 100 mg SR Capsules have not been studied in children, so their safety and efficacy in pediatric patients remain unestablished.
