Description
Indications
Anfree 0.5 mg Tablet, containing Flupentixol and Melitracen, is prescribed for the treatment of:
- Anxiety
- Depression
- Apathy
- Psychogenic depression
- Depressive neuroses
- Masked depression
- Psychosomatic disorders accompanied by anxiety and apathy
- Menopausal depression
- Mood disturbances (dysphoria) and depression in individuals with alcohol or substance dependency

Pharmacology
Anfree 0.5 mg Tablet combines two well-established compounds:
- Flupentixol – A neuroleptic agent with inherent anxiolytic and antidepressant properties in low doses.
- Melitracen – A bipolar thymoleptic with stimulating effects at low doses.
Together, they create a formulation with antidepressant, anxiolytic, and activating effects. After oral administration, both Flupentixol and Melitracen reach peak plasma concentration within approximately 4 hours. The elimination half-life is around 35 hours for Flupentixol and 19 hours for Melitracen. Their combination does not significantly alter the pharmacokinetics of the individual components.
Dosage & Administration
Adults: Typically, 2 tablets per day, taken in the morning and at noon. In severe cases, the morning dose may be increased to 2 tablets.
Elderly patients: Recommended dose is 1 tablet in the morning.
Maintenance dose: Usually 1 tablet in the morning. If the patient experiences severe restlessness or insomnia, additional sedative therapy may be required during the acute phase.

Drug Interactions
- May enhance the effects of alcohol, barbiturates, and other central nervous system (CNS) depressants.
- Concurrent use with monoamine oxidase (MAO) inhibitors may lead to hypertensive crises.
- Neuroleptics and thymoleptics may reduce the antihypertensive effect of guanethidine and similar drugs.
- Thymoleptics may enhance the effects of adrenaline and noradrenaline.
Contraindications
This medication should not be used in the following conditions:
- Immediate recovery phase following a myocardial infarction
- Bundle-branch conduction defects
- Untreated narrow-angle glaucoma
- Acute intoxication from alcohol, barbiturates, or opiates
- Patients who have taken MAO inhibitors within the past two weeks
- Not recommended for highly excitable or overactive individuals, as its stimulating effects may intensify these symptoms
Side Effects
At the recommended dosage, side effects are rare but may include transient restlessness and insomnia.
Use in Pregnancy & Lactation
This medication should be avoided during pregnancy and breastfeeding unless prescribed by a physician.
Precautions & Warnings
- If the patient was previously on sedative tranquilizers, gradual discontinuation is advised.
Overdose Effects
Overdose symptoms are primarily due to Melitracen and may include anticholinergic effects. In rare cases, extrapyramidal symptoms caused by Flupentixol may appear.
- Treatment: Supportive and symptomatic care is necessary.
- Gastric lavage should be performed as soon as possible, and activated charcoal may be administered.
- Cardiovascular and respiratory support should be provided.
- Epinephrine (adrenaline) must not be used.
- Convulsions may be treated with Diazepam.
- Extrapyramidal symptoms can be managed with Biperiden.
