Description
Indications
Antrenex is prescribed for the management of various gastrointestinal and smooth muscle spasms. It is effective in conditions such as gastrointestinal hypermotility, gastroesophageal spasms, gastroduodenitis, spastic constipation, pylorospasm, and peptic ulcers. It is also used to relieve spasms and pain associated with carcinoma, biliary tract disorders, and urinary tract conditions.

Pharmacology
Oxyphenonium is an anticholinergic agent that works by inhibiting the action of acetylcholine, a neurotransmitter responsible for muscle stimulation. By blocking acetylcholine receptors in smooth muscles, it reduces muscle contractions and provides relief from spasms. Additionally, Oxyphenonium exerts a direct relaxing effect on smooth muscles, making it beneficial in conditions like irritable bowel syndrome. It also helps reduce gastrointestinal motility, decreases gastric acid secretion, and controls excessive secretions in the pharynx, trachea, and bronchi.
Dosage & Administration
For adults, the usual dosage is 10 mg (2 tablets) taken four times daily for several days. The dose may be adjusted based on the patient’s response or as advised by the physician.

Drug Interactions
Oxyphenonium may reduce gastric motility, which can affect the absorption of other medications. Its effects may be enhanced when taken with other antimuscarinic agents such as amantadine, certain antihistamines, phenothiazines, and tricyclic antidepressants (TCA).
Contraindications
Antrenex should not be used in patients with the following conditions:
- Glaucoma
- Obstructive uropathy (e.g., bladder neck obstruction due to prostatic hypertrophy)
- Gastrointestinal obstruction (e.g., achalasia, paralytic ileus, pyloroduodenal stenosis)
- Intestinal atony in elderly or debilitated individuals
- Unstable cardiovascular status during acute hemorrhage
- Severe ulcerative colitis or toxic megacolon
- Myasthenia gravis
- Known hypersensitivity to the drug
Side Effects
Common side effects of Antrenex include:
- Dry mouth
- Headache
- Palpitations
Use in Pregnancy & Lactation
Animal studies have shown potential adverse effects on fetal development, but adequate studies in humans are not available. This medication should only be used during pregnancy if the potential benefits outweigh the possible risks to the fetus.
Precautions & Warnings
Special caution should be taken in patients with:
- Children and elderly individuals
- Benign prostatic hyperplasia
- Acute myocardial infarction (MI)
- Heart failure or hypertension
- Thyrotoxicosis
- Fever
- Angle-closure glaucoma
- Pregnancy and lactation
Overdose Management
The treatment of Oxyphenonium overdose follows the same approach as atropine toxicity management.
