Description
Indications:
Beklo is indicated for the management of various conditions, including:
- Spasticity resulting from multiple sclerosis
- Flexor spasms along with concomitant pain, clonus, and muscular rigidity
- Skeletal muscle spasms caused by rheumatic disorders
- Spinal cord injuries and other spinal cord diseases
- Cerebrovascular accidents, neoplastic diseases, or degenerative brain diseases

Pharmacology:
Beklo contains Baclofen, a medication that works by inhibiting both monosynaptic and polysynaptic reflexes in the spinal cord. It stimulates GABAB receptors, which in turn reduce the release of glutamate and aspartate, effectively relaxing the muscles. Baclofen also provides an antinociceptive effect, reducing pain sensations.
Dosage & Administration:
- For Adults and Children over 10 years: The starting dose is 5 mg three times daily, preferably with or after food. The dosage may be gradually increased up to a maximum of 100 mg daily.
- For Children under 10 years:
- Children aged 12 months to 2 years: Start with 10-20 mg (10-20 ml) per day.
- Children aged 2 to 6 years: Start with 20-30 mg (20-30 ml) per day.
- Children aged 6 to 10 years: Start with 30-60 mg (30-60 ml) per day.
- The medication should be increased gradually as required by the patient’s condition.

Interactions:
- CNS depressants: Increased sedation may occur when Beklo is taken with alcohol, synthetic opiates, or other CNS depressants, raising the risk of respiratory depression.
- Antihypertensive drugs: When combined with Beklo, antihypertensive medication may result in an enhanced drop in blood pressure, necessitating possible dose adjustments.
- Tricyclic antidepressants: The combination of Beklo with tricyclic antidepressants can amplify the muscle-relaxing effects of Beklo, causing pronounced muscle hypotonia.
- Parkinson’s disease treatments: Co-administration with levodopa and carbidopa may lead to mental confusion, hallucinations, headaches, nausea, and agitation.
- MAO inhibitors: The use of Beklo with MAO inhibitors may increase CNS depressant effects. Dosing adjustments may be needed.
- Neuromuscular blocking agents: Caution should be exercised when Beklo is used in conjunction with magnesium sulfate or other neuromuscular blocking agents due to a possible synergistic effect.
Contraindications:
Beklo is contraindicated for patients with a known hypersensitivity to any of its components.
Side Effects:
The most common side effects include transient drowsiness, daytime sedation, dizziness, weakness, and fatigue. Other side effects may include:
- CNS-related: Headache, insomnia, confusion, hallucinations, tremors, muscle pain, and seizures.
- Cardiovascular: Hypotension, dyspnea, palpitation, chest pain, and syncope.
- Gastrointestinal: Nausea, constipation, dry mouth, abdominal pain, anorexia, and vomiting.
- Genitourinary: Increased urinary frequency, urinary retention, dysuria, impotence, and hematuria.
- Other: Rash, excessive perspiration, weight gain, visual disturbances, and liver function disorders.
Pregnancy & Lactation:
Pregnancy Category B3. Baclofen should only be used during pregnancy when the benefits outweigh the risks. It crosses the placental barrier, and safety during pregnancy has not been established. Baclofen is also excreted in breast milk, but the quantities are considered too low to affect the infant. Use caution when administering to breastfeeding women.
Precautions & Warnings:
- Renal impairment: Lower doses (around 5 mg per day) should be used for patients with kidney issues or those undergoing chronic hemodialysis.
- Psychiatric disorders: Use cautiously in patients with psychiatric conditions such as schizophrenia, depression, or psychosis, as these disorders may worsen.
- Epilepsy: In patients with both epilepsy and muscle spasticity, Beklo can be used cautiously, but adequate anticonvulsive therapy should be maintained.
- Peptic ulcers and cardiovascular conditions: Caution is advised for patients with peptic ulcers, cerebrovascular diseases, or a history of cardiovascular conditions.
- Respiratory function: Close monitoring of respiratory and cardiovascular function is necessary, especially in patients with respiratory muscle weakness or cardiopulmonary diseases.
- Bladder function: Beklo may improve neurogenic bladder disturbances but should be used with caution in patients with existing sphincter hypertonia.
Overdose Effects:
In cases of severe overdose, gastric lavage may be necessary. Seek immediate medical attention in such situations.
Therapeutic Class:
Beklo belongs to the class of centrally acting skeletal muscle relaxants.
Storage Conditions:
Store below 30°C, away from light and moisture. Keep out of reach of children.
