Indications
Clarizol is indicated for the treatment of dermatomycoses caused by various fungal pathogens, including:

  • Dermatophyte infections (e.g., Trichophyton species)
  • Yeast infections (e.g., Candida species)
  • Mould-induced dermatomycoses and other fungal skin conditions
  • Skin infections with superimposed fungal infections

Common conditions treated include interdigital mycoses (e.g., athlete’s foot), paronychias (associated with nail mycoses), skin fold mycoses, Candida vulvitis, Candida balanitis, Pityriasis versicolor, and erythrasma.

Description
Clarizol is a broad-spectrum antifungal agent designed to treat dermal infections caused by dermatophytes, yeasts, moulds, and certain gram-positive bacteria, particularly Staphylococci and Trichomonas. Like other imidazoles, Clarizol disrupts fungal cell membrane permeability by interfering with essential cell wall lipids. At low concentrations, it inhibits ergosterol synthesis, while at higher doses, it directly damages fungal cell membranes, leading to cell death.

Pharmacology
Clotrimazole, the active ingredient in Clarizol, exerts its antifungal effect by inhibiting ergosterol biosynthesis—an essential component of fungal cell membranes. This disruption compromises membrane integrity, preventing fungal cell growth and replication.

Additionally, Clotrimazole has other pharmacological effects, including:

  • Inhibition of sarcoplasmic reticulum Ca2+ ATPase
  • Depletion of intracellular calcium
  • Blocking calcium-dependent potassium channels and voltage-dependent calcium channels

These secondary effects contribute to its broad-spectrum antifungal activity.

Dosage & Administration

  • Clotrimazole Cream: Apply a thin layer to the affected area 2-3 times daily and gently rub in. Due to its high efficacy, a small amount is usually sufficient to cover an area the size of a palm.
  • Recommended treatment duration:
    • Dermatomycoses: 3-4 weeks
    • Candida vulvitis and Candida balanitis: 1-2 weeks
    • Erythrasma and Pityriasis versicolor: ~3 weeks
    • For foot infections, continue treatment for 2 weeks beyond symptom resolution to prevent recurrence. Ensure feet are thoroughly dried after washing, especially between toes.
  • Clotrimazole Topical Solution: Apply sparingly to affected areas and rub in gently, two or three times daily.

Interaction
No significant drug interactions have been reported.

Contraindications
Clarizol is contraindicated in individuals with known hypersensitivity to Clotrimazole.

Side Effects
Clarizol is generally well tolerated. When applied topically, systemic side effects are rare. In some cases, mild local irritation or a burning sensation may occur, but these effects are not considered harmful.

Pregnancy & Lactation
Clarizol should only be used during pregnancy if deemed necessary by a physician.
