Description
Indications
Coralex D tablet is indicated for the treatment and prevention of:
- Osteoporosis
- Osteomalacia
- Tetany
- Hypoparathyroidism
- Disorders of osteogenesis
It is also used as a supplement in cases of inadequate calcium intake during childhood, rickets, pregnancy, lactation, and in elderly patients. Additional uses include treatment for pancreatitis and as a phosphate binder in chronic renal failure.
Use this medication only as directed by a registered physician.
Composition
Each tablet contains:
- Coralex D Tablet: Calcium Carbonate USP 1250 mg (Coral source) equivalent to 500 mg of elemental Calcium & Vitamin D USP 200 IU.
- Coralex D DX Tablet: Calcium Carbonate USP 1500 mg (Coral source) equivalent to 600 mg of elemental Calcium & Vitamin D USP 400 IU.
- Coralex D Effervescent Tablet: Calcium Carbonate (Coral source) USP 1500 mg equivalent to 600 mg of elemental Calcium and Vitamin D3 400 IU as Cholecalciferol USP.
Pharmacology
Calcium is essential for numerous bodily functions, and its deficiency can lead to serious health issues. Clinical studies demonstrate that Calcium aids in the prevention and treatment of osteoporosis and associated fractures. Vitamin D enhances calcium absorption from the gastrointestinal tract and supports bone formation. Coral-derived Calcium closely resembles human bone composition, ensuring better absorption. Vitamin D3 further helps maintain calcium balance in the body.
Dosage & Administration
- Tablet Form: Take one tablet once or twice daily with plenty of water, preferably with food for better absorption.
- Effervescent Tablet: Dissolve one tablet in a glass of water, stir, and drink. Best taken with food.

Drug Interactions
- Thiazide diuretics reduce calcium excretion, increasing the risk of hypercalcemia.
- Systemic corticosteroids may lower calcium absorption, necessitating dosage adjustments.
- Cholestyramine and paraffin oil may impair Vitamin D absorption.
- Calcium may reduce tetracycline absorption; administer tetracycline at least two hours before or four to six hours after calcium intake.
- Hypercalcemia may heighten the toxicity of cardiac glycosides; ECG and calcium levels should be monitored.
- Bisphosphonates or sodium fluoride should be taken at least three hours before calcium intake.
- Foods rich in oxalic acid (e.g., spinach) or phytic acid (e.g., whole cereals) may inhibit calcium absorption. Avoid taking calcium within two hours of such foods.
Contraindications
Do not use Coralex D in cases of:
- Hypercalcemia or hyperparathyroidism
- Hypercalciuria or kidney stones
- Severe renal insufficiency
- Hypersensitivity to any component
- Concomitant digoxin therapy (requires careful calcium level monitoring)
Side Effects
- Gastrointestinal irritation, constipation, nausea, vomiting
- Hypercalcemia (rare but possible in patients with chronic renal failure)
- Allergic reactions, irregular heartbeat, dry mouth, and drowsiness
- Occasional skin rash due to Vitamin D supplementation
Pregnancy & Lactation
- Daily intake should not exceed 1500 mg Calcium and 600 IU Vitamin D.
- High doses of Vitamin D have shown reproductive toxicity in animal studies.
- Avoid overdoses as hypercalcemia may negatively impact fetal development.
- Calcium and Vitamin D pass into breast milk, requiring consideration when supplementing Vitamin D in infants.
Precautions & Warnings
- Long-term treatment requires monitoring of serum calcium levels and renal function.
- Elderly patients and those on cardiac glycosides or diuretics require special monitoring.
- Patients with renal impairment should use Vitamin D cautiously to prevent soft tissue calcification.
- Patients with sarcoidosis require calcium level monitoring due to potential Vitamin D metabolism abnormalities.
- Immobilized patients with osteoporosis have a higher risk of hypercalcemia.
- Avoid additional Vitamin D supplementation unless medically supervised.
Overdose Effects
Symptoms of overdose include:
- Anorexia, thirst, nausea, vomiting, constipation, abdominal pain
- Muscle weakness, fatigue, mental disturbances, excessive urination
- Severe cases may result in cardiac arrhythmias, coma, or death
Management of Overdose:
- Discontinue calcium and Vitamin D intake.
- Stop concurrent use of thiazide diuretics, lithium, Vitamin A, and cardiac glycosides.
- Gastric lavage for unconscious patients.
- Rehydration and treatment with loop diuretics, bisphosphonates, calcitonin, and corticosteroids as necessary.
- Monitor serum electrolytes, renal function, and diuresis.
